hover_enabled=&#8221;0&#8243; global_colors_info=&#8221;{}&#8221; sticky_enabled=&#8221;0&#8243;]\n<p class=\"ai-optimize-60\"><span style=\"font-weight: 400;\">New research has demonstrated the significant role stem cells play in skin and hair development, which could lead to revolutionary new anti-ageing treatments. <\/span><a href=\"https:\/\/www.bbc.co.uk\/news\/articles\/c756q166gd9o\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[1]<\/span><\/a><\/p>\n<p class=\"ai-optimize-61\"><span style=\"font-weight: 400;\">By identifying the ways in which the body grows skin, researchers believe that their findings could lead to the use of artificial skin in treatments for burns, wounds, and age-related degeneration.<\/span><\/p>\n<p class=\"ai-optimize-62\"><span style=\"font-weight: 400;\">The research was conducted as part of <\/span><a href=\"https:\/\/www.humancellatlas.org\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">The Human Cell Atlas project<\/span><\/a><span style=\"font-weight: 400;\">, which aims to map the genetic instructions contained in every cell in the human body to discover how they work.\u00a0<\/span><\/p>\n<p>&nbsp;<\/p>\n<h3 class=\"ai-optimize-63\"><span style=\"font-weight: 400;\">New discoveries about skin formation<\/span><\/h3>\n<p class=\"ai-optimize-64\"><span style=\"font-weight: 400;\">The study looked at the role immune cells, specifically macrophages, play in the development of skin during pregnancy.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-65\"><span style=\"font-weight: 400;\">While macrophages &#8211; which are a type of white blood cell responsible for killing pathogens &#8211; are primarily known for their importance as part of the immune system, researchers sought to identify how they contribute to skin formation in a process known as morphogenesis.<\/span><\/p>\n<p class=\"ai-optimize-66\"><span style=\"font-weight: 400;\">In order to do this, a so-called \u2018reference atlas\u2019 of prenatal skin (that is, skin that forms between 7-17 weeks after conception) was made using advanced techniques including single cell and spatial transcriptomics &#8211; a study of RNA molecules within both one and a population of cells &#8211; to detail how elements both within and between skin cells interact.<\/span><\/p>\n<p class=\"ai-optimize-67\"><span style=\"font-weight: 400;\">Their findings showed that interactions between immune cells like macrophages and non-immune cells were crucial for key processes in morphogenesis, including hair follicle development, scarless wound healing, and the formation of blood vessels in the skin. <\/span><a href=\"https:\/\/doi.org\/10.1038\/s41586-024-08002-x\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[2]<\/span><\/a><\/p>\n<p>&nbsp;<\/p>\n<h3 class=\"ai-optimize-68\"><span style=\"font-weight: 400;\">The important role of stem cells in determining skin formation\u00a0<\/span><\/h3>\n<p class=\"ai-optimize-69\"><span style=\"font-weight: 400;\">Researchers were able to verify their findings about the significant role played by immune cells in skin morphogenesis by comparing prenatal and adult skin with a lab-grown skin model derived from stem cells.<\/span><\/p>\n<p class=\"ai-optimize-70\"><span style=\"font-weight: 400;\">The model, known as an organoid, successfully mimicked many aspects of skin and hair follicle development found in prenatal and adult skin.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-71\"><span style=\"font-weight: 400;\">However, it was only when researchers added macrophages that they found the model created blood vessel networks, confirming that immune cells play a more significant role in the development of skin than previously thought.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-72\"><span style=\"font-weight: 400;\">This stem-cell derived model of human skin is thought to have a vast array of possible therapeutic applications extending from use in identifying the development of conditions affecting skin and hair to providing a more accurate test material for future skin and hair treatments. <\/span><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC7593871\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[3]<\/span><\/a><\/p>\n<p>&nbsp;<\/p>\n<h3 class=\"ai-optimize-73\"><span style=\"font-weight: 400;\">Mesenchymal stem cells &#8211; key players in hair and skin formation<\/span><\/h3>\n<p class=\"ai-optimize-74\"><span style=\"font-weight: 400;\">Another area researchers looked at during the study was the formation of hair follicles during prenatal development.<\/span><\/p>\n<p class=\"ai-optimize-75\"><span style=\"font-weight: 400;\">In earlier studies with mice, researchers identified special cells called pre-Dc cells that eventually become part of the hair follicle structure (called the dermal condensate, or Dc). <\/span><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC9504245\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[4]<\/span><\/a><\/p>\n<p class=\"ai-optimize-76\"><span style=\"font-weight: 400;\">Researchers identified similar cells in human prenatal skin and tracked how they developed into the Dc and another structure called the dermal papilla (Dp), which is important for hair growth.<\/span><\/p>\n<p class=\"ai-optimize-77\"><span style=\"font-weight: 400;\">Using various advanced methods, researchers were able to map out the development of these cells over time and predict their paths.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-78\"><span style=\"font-weight: 400;\">They found that certain early skin cells (fibroblasts) go down one of two routes: some become specialized for hair follicle formation, while others develop into more general skin cells.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-79\"><span style=\"font-weight: 400;\">As the hair-specific cells move toward the outer layer of the skin, they turn on genes that help them stick together and form the structures needed for hair growth.<\/span><\/p>\n<p class=\"ai-optimize-80\"><span style=\"font-weight: 400;\">The study also looked at how mesenchymal cells interact with skin\u2019s outer (epithelial) cells during hair follicle formation.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-81\"><span style=\"font-weight: 400;\">They identified important signalling molecules, such as CXCL12, that help guide cell migration and organization. Interestingly, immune cells were also predicted to assist with these processes. <\/span><a href=\"https:\/\/www.nature.com\/articles\/s41586-024-08002-x\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[5]<\/span><\/a><\/p>\n<p>&nbsp;<\/p>\n<h3 class=\"ai-optimize-82\"><span style=\"font-weight: 400;\">Discovering the genetic origins of skin and hair conditions<\/span><\/h3>\n<p class=\"ai-optimize-83\"><span style=\"font-weight: 400;\">Having mapped how skin and hair developed in prenatal skin, researchers turned to investigating the origins of certain skin and hair conditions.<\/span><\/p>\n<p class=\"ai-optimize-84\"><span style=\"font-weight: 400;\">They found that reduced hair growth (<\/span><a href=\"https:\/\/www.cancer.gov\/publications\/dictionaries\/cancer-terms\/def\/hypotrichosis\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">hypotrichosis<\/span><\/a><span style=\"font-weight: 400;\">) and abnormal hair shapes (like <\/span><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC8432236\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">pili torti<\/span><\/a><span style=\"font-weight: 400;\">) are caused by specific genes that are active during early hair follicle development.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-85\"><span style=\"font-weight: 400;\">Similarly, they were able to identify that <\/span><a href=\"https:\/\/www.nhs.uk\/conditions\/epidermolysis-bullosa\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">epidermolysis bullosa (EB)<\/span><\/a><span style=\"font-weight: 400;\"> &#8211; a disorder that makes the skin fragile and prone to blistering &#8211; had its roots in the expression of the gene COL7A1 in several types of prenatal skin cells.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-86\"><span style=\"font-weight: 400;\">Researchers were again able to compare gene expression in prenatal skin with their stem cell-derived lab-grown skin models and found strong similarities, noting that while these diseases often stem from defects in skin structure, they are frequently worsened by immune system involvement. <\/span><a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC9290674\/\" target=\"_blank\" rel=\"noreferrer noopener\"><span style=\"font-weight: 400;\">[6]<\/span><\/a><\/p>\n<p>&nbsp;<\/p>\n<h3 class=\"ai-optimize-87\"><span style=\"font-weight: 400;\">The importance of stem cells and cord blood banking<\/span><\/h3>\n<p class=\"ai-optimize-88\"><span style=\"font-weight: 400;\">As this new research demonstrates, stem cells play a pivotal role in not only helping researchers to better understand the formation of vital organs, like skin, but also in helping to facilitate potential future therapies.\u00a0<\/span><\/p>\n<p class=\"ai-optimize-89\"><span style=\"font-weight: 400;\">By being able to compare cells from both prenatal and adult skin with artificial lab-grown skin using stem cells, researchers were able to identify the previously unknown significance of immune cells in skin formation and accurately map the genetic instructions involved in growing skin.<\/span><\/p>\n<p class=\"ai-optimize-90\"><span style=\"font-weight: 400;\">In their paper published in Nature, The Human Cell Atlas team have achieved something remarkable: essentially providing a set of instructions for creating human skin.<\/span><\/p>\n<p class=\"ai-optimize-91\"><span style=\"font-weight: 400;\">Their research is being heralded as groundbreaking and for good reason. But when feeling down persists for weeks, or even months, it can be a sign of a mental health condition, like depression.<\/p><p>Although today people are well versed and more understanding of depression, some sufferers still find it difficult to find treatments.<\/p><h3>What is depression?<\/h3><p>Depression is a common mental health condition. It is characterised by low mood, feelings of hopelessness, losing interest in things which previously brought a sense of enjoyment, and feeling tearful.<\/p><p>Depression can also manifest in physical ways. Sleeping poorly, feeling tired, experiencing aches, pains, a diminished appetite, and decreased libido can all be impacts of depression. The symptoms range from mild to severe. In mild cases, a persistent low mood may be the cause, while severe cases may result in suicidal feelings or a desire to self harm <a href=\"https:\/\/www.nhs.uk\/mental-health\/conditions\/depression-in-adults\/overview\/\">[1]<\/a>.<\/p><p>Sometimes the causes of depression can be obvious, such as a bereavement or a traumatic event. Other times there might be no known cause, although family history could be the reason. Whatever the reason, once a person has identified they may be depressed, they should go and see their GP.<\/p><h3>What treatments are available?<\/h3><p>There are a plethora of treatments for depression that a doctor can prescribe, from talking therapies to pharmaceutical medications to exercise and more. Sometimes the GP may recommend one treatment or a combination of treatments but they will help support patients and signpost them to services which can bolster their support network.<\/p><p>The good news is that depression is among the most treatable mental health disorders.\u00a0 Between 80% and 90% percent of people with depression eventually respond well to treatment. Almost all patients gain some relief from their symptoms <a href=\"https:\/\/www.psychiatry.org\/patients-families\/depression\/what-is-depression#:~:text=Depression%20is%20among%20the%20most,some%20relief%20from%20their%20symptoms.\">[2]<\/a>.<\/p><h3>Treatment Resistant Depression<\/h3><p>For those with major depressive disorder, however, treatment can be harder to find. With up to one third of adults with the condition failing to respond to treatment, new avenues and treatments are being sought.<\/p><p>\u201cAlthough there is some disagreement as to how to define treatment-resistant depression, a patient is generally considered to have it if the individual hasn\u2019t responded to adequate doses of two different antidepressants taken for a sufficient duration of time, which is usually six weeks,\u201d explains Jaskaran Singh, M.D., Senior Director of Neuroscience, Janssen Pharmaceuticals <a href=\"https:\/\/www.jnj.com\/health-and-wellness\/4-facts-about-treatment-resistant-depression\">[3]<\/a>.<\/p><p>The theory that depression may have causes currently unknown by scientists is one theory researchers are exploring to try and combat resistance to treatment.<\/p><p>\u201cOne of the more modern theories is that depression creates inflammation in the brain, or that inflammation in the brain creates depression. Traditional antidepressants only affect neurotransmitters, so this may be why some patients don\u2019t respond to them.\u201d\u00a0 says Alexander Papp, M.D., a psychiatrist at UC San Diego Health <a href=\"https:\/\/www.jnj.com\/health-and-wellness\/4-facts-about-treatment-resistant-depression\">[4]<\/a>.<\/p><p>Whether or not this is the case, researchers are still trying to find solutions to Treatment Resistant Depression.<\/p><h3>The Potential of Cord Blood Therapy<\/h3><p>One study using umbilical cord blood cells delivered promising results. The study administered concentrated umbilical cord blood cells at a dose of 250 million cells via weekly injections over 4 weeks with 1 week intervals, to women who suffered with Treatment Resistant Depression.\u00a0 The results were tested against a control group who received a placebo.<\/p><p>The study found that the therapeutic potential of umbilical cord blood cell concentrate can be used to overcome treatment resistance formed in depressive patients <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7087609\/\">[5]<\/a>. Other studies have also found that cellular therapies may have a role in combating Treatment Resistant Depression <a href=\"https:\/\/www.mdpi.com\/1999-4923\/15\/3\/814\">[6]<\/a> <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC8474995\/\">[7]<\/a>.<\/p><p>These studies give hope for finding new treatments.
