{"id":38628,"date":"2025-08-26T15:29:06","date_gmt":"2025-08-26T15:29:06","guid":{"rendered":"https:\/\/cells4life.com\/us\/?p=38628"},"modified":"2025-09-29T16:06:04","modified_gmt":"2025-09-29T16:06:04","slug":"stem-cell-treatment-could-help-newborns-with-brain-damage","status":"publish","type":"post","link":"https:\/\/cells4life.com\/us\/2025\/08\/stem-cell-treatment-could-help-newborns-with-brain-damage\/","title":{"rendered":"Stem Cell Treatment Could Help Newborns With Brain Damage"},"content":{"rendered":"[et_pb_section fb_built=&#8221;1&#8243; _builder_version=&#8221;4.16&#8243; global_colors_info=&#8221;{}&#8221;][et_pb_row _builder_version=&#8221;4.16&#8243; background_size=&#8221;initial&#8221; background_position=&#8221;top_left&#8221; background_repeat=&#8221;repeat&#8221; global_colors_info=&#8221;{}&#8221;][et_pb_column type=&#8221;4_4&#8243; _builder_version=&#8221;4.16&#8243; custom_padding=&#8221;|||&#8221; global_colors_info=&#8221;{}&#8221; custom_padding__hover=&#8221;|||&#8221;][et_pb_text _builder_version=&#8221;4.27.4&#8243; background_size=&#8221;initial&#8221; background_position=&#8221;top_left&#8221; background_repeat=&#8221;repeat&#8221; locked=&#8221;off&#8221; global_colors_info=&#8221;{}&#8221;]\n<p class=\"ai-optimize-14\" style=\"font-weight: 400;\">A recent study conducted at UMC Utrecht, Netherlands, has shown that mesenchymal stem cells (MSCs) are a safe treatment for brain damage caused by stroke in newborns, and could have a significant positive effect.<\/p>\n<h3 class=\"ai-optimize-15\"><span style=\"font-weight: 400;\">Perinatal stroke and its consequences<\/span><\/h3>\n<p class=\"ai-optimize-16\" style=\"font-weight: 400;\">Perinatal stroke is defined as a stroke that happens to unborn or newborn babies before, during, or shortly after birth, typically between the last few months of pregnancy and one month of age.<a href=\"https:\/\/www.stroke.org\/en\/about-stroke\/stroke-in-children\/perinatal-stroke-infographic\" target=\"_blank\" rel=\"noopener\">[1]<\/a> It is estimated to affect about 1 in 1000 to 3000 children at birth[1][2], although the real figure may be higher as it is likely underdiagnosed.<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/B9780444642394000163\" target=\"_blank\" rel=\"noopener\">[2]<\/a><\/p>\n<p class=\"ai-optimize-17\" style=\"font-weight: 400;\">Perinatal stroke, particularly perinatal arterial ischemic stroke (PAIS) which is the most common type (70% of cases)<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/B9780444642394000163\" target=\"_blank\" rel=\"noopener\">[2]<\/a>, can have serious consequences. Depending on the artery involved, 50-70% of children who suffered a perinatal stroke will have lifelong issues, including cerebral palsy, epilepsy, vision issues, cognitive impairment and language delays.<a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">[3]<\/a> There is currently no treatment available that can alleviate these issues; the current standard of care is limited to supportive measures.<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/B9780444642394000163\" target=\"_blank\" rel=\"noopener\">[2]<\/a><a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">[3]<\/a> Thus, there is an urgent need to develop an effective regenerative treatment and ensure children who suffered a stroke get the best possible start in life.<\/p>\n<h3 class=\"ai-optimize-18\"><span style=\"font-weight: 400;\">The PASSIoN study<\/span><\/h3>\n<p class=\"ai-optimize-19\" style=\"font-weight: 400;\">A research team at UMC Utrecht has been focusing on developing a treatment for perinatal stroke using MSCs.<a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">[3]<\/a> These powerful stem cells offer several advantages, boasting neuroregenerative and anti-inflammatory properties that could prove key to reducing the impact of stroke on a newborn\u2019s brain.<a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/dmcn.13528\" target=\"_blank\" rel=\"noopener\">[4]<\/a> Administration of the MSCs via nose drops was selected as being the method of treatment that could potentially deliver the most stem cells to the brain.<a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">[3]<\/a><a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/dmcn.13528\" target=\"_blank\" rel=\"noopener\">[4]<\/a><\/p>\n<p class=\"ai-optimize-20\" style=\"font-weight: 400;\">Following on from preclinical studies in mice, researchers set up a phase 1 clinical trial, called PASSIoN (Perinatal Arterial Stroke treated with Stromal cells IntraNasally), to determine the safety of the treatment.<\/p>\n<p class=\"ai-optimize-21\" style=\"font-weight: 400;\">Newborns were eligible for inclusion into the trial if they were born at any of the neonatal intensive care units in the Netherlands and showed symptoms of PAIS, including seizures and breathing difficulties. Eligible newborns were transferred to UMC Utrecht, where an MRI was performed to confirm whether they had a stroke; if so, they received the stem cell treatment within 7 days after first showing symptoms. In total, ten newborns received the treatment between February 2020 and April 2021.<a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">[3]<\/a><\/p>\n<h3 class=\"ai-optimize-22\"><span style=\"font-weight: 400;\">What were the results?<\/span><\/h3>\n<p class=\"ai-optimize-23\" style=\"font-weight: 400;\">Initial results for the study were published in 2022, in the prestigious <a href=\"https:\/\/www.thelancet.com\/journals\/laneur\/article\/PIIS1474-4422(22)00117-X\/abstract\" target=\"_blank\" rel=\"noopener\">The Lancet journal<\/a>. The treatment proved to be safe, with no serious adverse events found either immediately after treatment or at the three-month follow-up visit. One newborn did develop a high fever after treatment, but it passed spontaneously within a few hours. Moreover, an MRI scan done at the three-month follow-up showed no unexpected brain abnormalities.<\/p>\n<p class=\"ai-optimize-24\" style=\"font-weight: 400;\">More recently, researchers published long-term follow-up results for the study, along with a comparison between the study patients and a cohort of newborns with stroke who received standard of care treatment. This comparison cohort was selected from the Neonatal Stroke Registry Utrecht using criteria that would have resulted in their enrollment in the PASSIoN trial had it been running at the time of their birth.<a href=\"https:\/\/www.ahajournals.org\/doi\/10.1161\/STROKEAHA.125.050786\" target=\"_blank\" rel=\"noopener\">[5]<\/a><a href=\"https:\/\/research.umcutrecht.nl\/news\/stem-cell-treatment-offers-hope-for-newborns-with-brain-damage\/\" target=\"_blank\" rel=\"noopener\">[6]<\/a><\/p>\n<p class=\"ai-optimize-25\" style=\"font-weight: 400;\">Two years after treatment, the children continue to experience no side effects; although there were two hospital admissions among the study group, these were found to have been unrelated to the treatment. Most children developed well from a cognitive point of view, with one showing a mild cognitive delay, two having language delays, and one suffering from severe sleep problems. MRI scans showed that the amount of brain tissue loss was less than would have been expected given the severity of the strokes.<\/p>\n<p class=\"ai-optimize-26\" style=\"font-weight: 400;\">In terms of motor development, too, the children who received the treatment performed better than the comparison group. All children in the study initially showed damage to the areas of the brain that control movement. This is something which usually carries a risk of developing cerebral palsy higher than 80%. However, only two children (20%) in the study group developed mild CP. In the comparison group, the rate of CP was 50%, and scientific literature reports rates of up to 70%.<\/p>\n<h3 class=\"ai-optimize-27\"><span style=\"font-weight: 400;\">A long-awaited paradigm shift<\/span><\/h3>\n<p class=\"ai-optimize-28\" style=\"font-weight: 400;\">Pediatrician and professor Manon Benders from UMC Utrecht, who co-led the PASSIoN study, says that seeing this kind of positive development in such a high-risk group is truly extraordinary.<a href=\"https:\/\/research.umcutrecht.nl\/news\/stem-cell-treatment-offers-hope-for-newborns-with-brain-damage\/\" target=\"_blank\" rel=\"noopener\">[6]<\/a><\/p>\n<p class=\"ai-optimize-29\" style=\"font-weight: 400;\">Following the success of this safety study, a larger trial, called iSTOP-CP, is now due to begin in early 2026. This trial will include a total of 162 newborns with brain damage due to either perinatal stroke or severe oxygen deprivation. Researchers will administer either the stem cell treatment or a placebo within 7 days of birth, then monitor their development up to the age of 24 months. It is hoped that the larger study will confirm the treatment\u2019s effectiveness, bringing about a long-awaited paradigm shift in the treatment of neonatal brain damage.<a href=\"https:\/\/research.umcutrecht.nl\/news\/stem-cell-treatment-offers-hope-for-newborns-with-brain-damage\/\" target=\"_blank\" rel=\"noopener\">[6]<\/a><a href=\"https:\/\/www.ahajournals.org\/doi\/10.1161\/STROKEAHA.125.052219\" target=\"_blank\" rel=\"noopener\">[7]<\/a><\/p>\n<p class=\"ai-optimize-30\" style=\"font-weight: 400;\">Clinical trials and studies like this make it clear that stem cells could hold the key to treating many illnesses and conditions that currently have no cure. Indeed, a recent review article has statistically confirmed that infusions of cord blood, which is rich in young, potent stem cells, can be an <a href=\"https:\/\/cells4life.com\/us\/2025\/04\/evidence-supports-cord-blood-therapy-for-cerebral-palsy\/\">effective treatment to improve motor skills in children with CP<\/a>. In the case of neonatal brain damage, too, it could be possible to develop a treatment using the baby\u2019s own stem cells, collected from the cord blood.<a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/dmcn.13528\" target=\"_blank\" rel=\"noopener\">[4]<\/a><\/p>\n<p class=\"ai-optimize-31\" style=\"font-weight: 400;\">This would only be feasible, however, if the cord blood is collected and stored after birth. But when feeling down persists for weeks, or even months, it can be a sign of a mental health condition, like depression.<\/p><p>Although today people are well versed and more understanding of depression, some sufferers still find it difficult to find treatments.<\/p><h3>What is depression?<\/h3><p>Depression is a common mental health condition. It is characterised by low mood, feelings of hopelessness, losing interest in things which previously brought a sense of enjoyment, and feeling tearful.<\/p><p>Depression can also manifest in physical ways. Sleeping poorly, feeling tired, experiencing aches, pains, a diminished appetite, and decreased libido can all be impacts of depression. The symptoms range from mild to severe. In mild cases, a persistent low mood may be the cause, while severe cases may result in suicidal feelings or a desire to self harm <a href=\"https:\/\/www.nhs.uk\/mental-health\/conditions\/depression-in-adults\/overview\/\">[1]<\/a>.<\/p><p>Sometimes the causes of depression can be obvious, such as a bereavement or a traumatic event. Other times there might be no known cause, although family history could be the reason. Whatever the reason, once a person has identified they may be depressed, they should go and see their GP.<\/p><h3>What treatments are available?<\/h3><p>There are a plethora of treatments for depression that a doctor can prescribe, from talking therapies to pharmaceutical medications to exercise and more. Sometimes the GP may recommend one treatment or a combination of treatments but they will help support patients and signpost them to services which can bolster their support network.<\/p><p>The good news is that depression is among the most treatable mental health disorders.\u00a0 Between 80% and 90% percent of people with depression eventually respond well to treatment. Almost all patients gain some relief from their symptoms <a href=\"https:\/\/www.psychiatry.org\/patients-families\/depression\/what-is-depression#:~:text=Depression%20is%20among%20the%20most,some%20relief%20from%20their%20symptoms.\">[2]<\/a>.<\/p><h3>Treatment Resistant Depression<\/h3><p>For those with major depressive disorder, however, treatment can be harder to find. With up to one third of adults with the condition failing to respond to treatment, new avenues and treatments are being sought.<\/p><p>\u201cAlthough there is some disagreement as to how to define treatment-resistant depression, a patient is generally considered to have it if the individual hasn\u2019t responded to adequate doses of two different antidepressants taken for a sufficient duration of time, which is usually six weeks,\u201d explains Jaskaran Singh, M.D., Senior Director of Neuroscience, Janssen Pharmaceuticals <a href=\"https:\/\/www.jnj.com\/health-and-wellness\/4-facts-about-treatment-resistant-depression\">[3]<\/a>.<\/p><p>The theory that depression may have causes currently unknown by scientists is one theory researchers are exploring to try and combat resistance to treatment.<\/p><p>\u201cOne of the more modern theories is that depression creates inflammation in the brain, or that inflammation in the brain creates depression. Traditional antidepressants only affect neurotransmitters, so this may be why some patients don\u2019t respond to them.\u201d\u00a0 says Alexander Papp, M.D., a psychiatrist at UC San Diego Health <a href=\"https:\/\/www.jnj.com\/health-and-wellness\/4-facts-about-treatment-resistant-depression\">[4]<\/a>.<\/p><p>Whether or not this is the case, researchers are still trying to find solutions to Treatment Resistant Depression.<\/p><h3>The Potential of Cord Blood Therapy<\/h3><p>One study using umbilical cord blood cells delivered promising results. The study administered concentrated umbilical cord blood cells at a dose of 250 million cells via weekly injections over 4 weeks with 1 week intervals, to women who suffered with Treatment Resistant Depression.\u00a0 The results were tested against a control group who received a placebo.<\/p><p>The study found that the therapeutic potential of umbilical cord blood cell concentrate can be used to overcome treatment resistance formed in depressive patients <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7087609\/\">[5]<\/a>. Other studies have also found that cellular therapies may have a role in combating Treatment Resistant Depression <a href=\"https:\/\/www.mdpi.com\/1999-4923\/15\/3\/814\">[6]<\/a> <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC8474995\/\">[7]<\/a>.<\/p><p>These studies give hope for finding new treatments.
